Hi,

I'm looking at creating a master topical solution to maintain my current hair. I would really appreciate the input from experience members on dosage (per compound) and how I can create the most efficient topical vehicle.

Anti-androgen
- Enzalutamide

5AR Inhibitor
- Zinc Sulphate
- Vitamin B6 p5p

Estrogen (ER-beta stimulation)
- Estriol (preferably powder form)

Activate SHH, Wnt pathway, growth factors, down regulates TGF and inhibits 5AR
- Costunolide

I have used Enzalutamide previously with 0 side effects and maintained with little to no shedding for the last 9 months. However, I have recently run out and my next batch is due in a couple of days so it gives me the opportunity to add to my topical routine.

Enzalutamide stops any androgen from binding to a given receptor, entering the cell nucleus and prevents transcription of hair loss gene responses.

The 5AR inhibitor compounds have been selected as they are the primary actives within Zix and have been proven to lower DHT in the scalp. They inhibit 5AR (unsure on 1 or 2) which turns testosterone into DHT.

Estriol has also been selected as it is an alpha Estrogen receptor inhibitor, promoting beta Estrogen receptor activity and boosting hair growth signals. Apparently this is a POM compound though (Prescription Only Medication) so i'd like to know how other people get hold of this and potentially in powder form. Costonulide was tested on human hair follicle dermal papilla cells, and found that it decreased 5AR activity by 50%, significantly decreased transforming growth factor and other gene markers involved in Androgenetic Alopecia, increased Wnt activity, sonic hedgehog pathway, and cell proliferation viability. It increased depth and diameter of hair transplanted onto mice. (Source - user: Georgie)

My only issue is with a required vehicle to ensure all these compounds do their required jobs. I have the following so far in mind:

DMSO
- Essentially for solubilising Enzalutamide and Costonulide , I believe Enzalutamide is soluble at up to 93mg/ml and Costonulide is around 10mg/ml so this is my route of choice for these compounds.

Ethanols/Alcohols
- This is where i'm a bit hazy, Zix uses Ethyl Alcohol in the original formula. I have currently using 96% alcohol from Polish Vodka.

I really need some help here from somebody that understands the different types of alcohols and ethanols and their advantages in this situation.

Synthetic Organic Compound
- So far I have ordered PEG 400 which is recommended in the original Enzalutamide thread, however I have tried Propylene Glycol (PG) which doesnt seem to have the same effect. Again, help here is needed to fill in my lack of knowledge.

I hope that this thread can act as the start of a new topical solution, I would appreciate anybody's support in piecing together how to form this.

Looks like a good start-you should think of adding a corticosteroid a few times a week. Dexamthasome is preferred since it increases aromatase expression in scalp. I haven't found a source for estriol powder so my topical contains estradiol (also minoxidil and dexamethasone). Estriol is over the counter in US but comes in creams rather powder. Keep us posted on a source for estriol powder. I'd like use that in place of estradiol.

As far as a single topical goes this isn’t going to work . Costonulide has such low solubility on its own that it won’t dissolve in an already semi saturated solution .

im very interested in this shh pathway stimulator as some study says it could be one of the key factor to hairneogenesis why not just try it once a week with dermaneedling if the solubility is bad .

Hi all,

Quick update,

I've having some issues with Zix
Zinc Sulphate and Vitamin B6 p5p are both insoluble in both DMSO and distilled water, which quite frankly seems ridiculous. If left standing for a couple of minutes all the powder coagulates at the bottom. Cant see a way forward for this, I have no idea how people use Zix. Desperately need help here.

Costunolide is too expensive
Xian Hao Xuan Bio Technology Co., Ltd have given me the following prices for the following quantities. This is not particularly sustainable.

FOB Price: USD198/g for 5g
Freight:USD40 to US by express

Estriol Procurement
Currently awaiting delivery of 1G Estriol Powder from Shanghai Huirui Chemical Technology via Alibaba.

Topical enza does nothing. Many have tried it.

Probably a good thing that you can’t get Costu, because it’s actually a telomerase inhibitor.

Those 5ARs are weak, but I mean, good luck.

Estradiol if anything will be doing the heavy lifting.
