I think this one is more promising than HM. it’s late preclinical so cross your fingers.

Won't be until 2007 or early 2008 until we see an IND from P&G

Until then we can only dream about.

Since it is well established that aberrant Hh pathway activity
in skin can lead to abnormal differentiation and various
follicular hyperplasias including BCC (Hahn et al, 1996;
Johnson et al, 1996; Dahmane et al, 1997; Oro et al, 1997;
Xie et al, 1998; Aszterbaum et al, 1999; Grachtchouk et al,
2000; Nilsson et al, 2000; Sheng et al, 2002), the effect of
Hh-agonist on differentiation in the skin was analyzed.
Seven-wk-old C57BL/6 male mice were either treated with
one topical dose of vehicle, Hh-agonist, or depilated to induce
anagen. Although hair follicles from vehicle-treated
skin were still in telogen 10 d after treatment (Fig 4A) both
depilated (Fig 4F) and Hh-agonist-treated hair follicles
(Fig 4K) were in anagen VI of the hair cycle. At this substage
of anagen, hair follicle size, proliferation, and hair
production are at their maximum.

The long-term effects of Hh-agonist treatment on proliferation
and differentiation in mouse skin were also examined.
Seven-wk-old mice were treated with either one
topical dose of vehicle or Hh-agonist to induce anagen.
Approximately 1 y later, skin from both groups was analyzed
by light microscopy and immunohistochemistry. Hair follicles
from both vehicle (Fig 5A)- and Hh-agonist (Fig 5B)-
treated skin were in telogen.

Sound to me like a super minoxidil. But correct me if I'm wrong these drugs were never tested on male pattern baldness follicles? That's a whole different game than healthy hair.

According to the authors this mouse animal strain has been the most intesnively studied for male pattern baldness. Or it currently is the most accurate animal we have of male pattern baldness. P&G uses the same animal strain and got the same results as curis did before they partnered for this program.

When you shave a mouse the hair doesn't grow back for a long time. Or it keeps it in the resting phase for some reason. As you can see by the pics above. Hence the agonist treated mice hair got kicked into the growth phase, while the other mice stayed in the resting phase.

Side effects to this treatment is the real question if the FDA will allow this Hh agonist treatment to work. It will definitly not be an over the couter medication.

Hedgehog,

Dont get me wrong my friend, I hope hedgehog agonists turn out to the cure for baldness, BUT

I'd feel a hell of alot better if this treatment were tried on balding apes because they have a androgenic loss of hair like humans do and share over 95% of genes with us. Bonoboos are our closest genetic relative if I remember correctly. Chimps lose hair, orangutans, macaques.

The thing about mice is that LOTS of things have very drammatic effects on them that last much longer than they do in humans. minoxidil would be a cure for baldness based on its results in mice...............but we know its not for us. The male hormone slowing the hair back down, the immune system attacking " something" about the miniaturizing follicle that eventually causes cell death in the lower follicle. Perhaps hedgehog could be a treatment that has to be applied once a year?

Like I said though man....................I hope its a cure as well as cloining being a cure. I want to see baldness eradicated for those who do not wish to be bald. Everyone who is not bald goes on about it being "insignifigant", but tell that to an 17 year old or a woman with alopecia areata in her prime years............its devastating for these people.

When the Hh is turned on it actually prevents the immune system from attacking the cell sorta of like cancer cells aren't seen by the immune system. However, it will be a once applied application and hopefully quick at that. I would guess application's would really depend on how fast your hairs fall back out and doesn't grow back.

It was orginally going to be used for chemo therapy patients. However, curis along with P&G thought they could bring it to the market for male pattern baldness.
