Low-Dose 17-β-Estradiol Cream for Vaginal Atrophy in a Cohort without Prolapse: Serum Levels and Vaginal Response Including Tissue Biomarkers Associated with Tissue Remodeling

Serum E2 levels (pg/mL) were unchanged from baseline (mean (SD)) 7.7 (3.3) to eight 9.7 (5.7) and sixteen 8.7 (5.8) (p=0.24) weeks. VMI (mean (SD)) improved from baseline 34.2 (18.3) to eight 56.7 (13.1) and sixteen 54.5 (11.3) (p<0.001) weeks with no difference between eight and sixteen weeks. Vaginal dryness (p=0.03) and itching (p=0.02) improved. Tissue biomarker levels did not change (TGF-β p=0.35, NFKB p=0.74, eNOS p=0.80, iNOS p=0.24, TSP p=0.80).

17b estradiol is very important for vaginal health for women as they age however due to the fear of developing breast cancer, many women choose to not use estradiol. This study assesed the effectiveness of estradiol creat at revitilizing the vaginal athrophy while also measuring serum estradiol levels. They found that serum estradiol levels did not increase

This means that it is possible to use 17b estradiol topically with minimal or no systemic absorption.
For those of us who believe that estrogen is a key factor in hair regrowth/maintenance, this is huge. I will be trying to get a hold of this estrogen cream they used in the study.

If anyone is interested in trying this out I recommend having a drug like Nolvadex or Raloxifene on hand just in case you get symptoms of gyno, you can stop the estrogen and use nolvadex for a few days and you'll be fine

How much would you apply? You intend to apply it on the scalp?

You would apply as much as 1gram of the cream per day on the scalp. That is the max dosage which was tested on this study to not raise serum levels and was also effective at healing the vaginal walls (this means that it will be more than enough for you follicles

I would recommend to start at 0.5g of the cream which would be 50 micrograms of estradiol, then up it to 1g if you dont get any side effects

That would be one application, which usually contains 0,5 mg of estriol.

Open a book and learn what lies under the layer of skin you see. It is impossible because you want something to be absorbed right under the skin but not enough to be absorbed systemically. It’s impossible and so are those ridiculous monkey studies you kids post regularly. This ncbi meme frenzy needs to stop at some point.

To be honest I very much doubt that there will be no systemic absorption. The creams are used by transitional male to female gender re-assigner's on their skin so it is meant to be absorbed through the skin. Maybe small amounts are tolerable but it will go systemic.

Maybe a very low dosage (say, just enough to apply on your scalp two times a day) would not give side effects and would counteract the rise on testosterone given by dutasteride?

if the dosage in the study was enough to hypertrophy the vaginal walls then it will be enough to have an effect on male pattern baldness, obv you cant use it by itself

Well, the study that you are referring to says that serum estradiol levels were unaltered, but it didn't say anything about levels of leutenizing hormone or systemic absorption. There is a good possibility that it was absorbed, raised the levels, affected the leutenizing hormone levels by getting detected in the hypothalamus, and eventually lowered the body's own production of estradiol. But of course, that's only my own hypothesis.
