[h=3]Abstract[/h]Prostaglandin E2 (PGE2) has been reported to modulate angiogenesis, the process of new blood vessel formation, by promoting proliferation, migration and tube formation of endothelial cells. Endothelial progenitor cells are known as a subset of circulating bone marrow mononuclear cells that have the capacity to differentiate into endothelial cells. However, the mechanism underlying the stimulatory effects of PGE2 and its specific receptors on bone marrow-derived cells (BMCs) in angiogenesis has not been fully characterized. Treatment with PGE2 significantly increased the differentiation and migration of BMCs. Also, the markers of differentiation to endothelial cells, CD31 and von Willebrand factor, and the genes associated with migration, matrix metalloproteinases 2 and 9, were significantly upregulated. This upregulation was abolished by dominant-negative AMP-activated protein kinase (AMPK) and AMPK inhibitor but not protein kinase, a inhibitor. As a functional consequence of differentiation and migration, the tube formation of BMCs was reinforced. Along with altered BMCs functions, phosphorylation and activation of AMPK and endothelial nitric oxide synthase, the target of activated AMPK, were both increased which could be blocked by EP4 blocking peptide and simulated by the agonist of EP4 but not EP1, EP2 or EP3. The pro-angiogenic role of PGE2 could be repressed by EP4 blocking peptide and retarded in EP4(+/-) mice. Therefore, by promoting the differentiation and migration of BMCs, PGE2 reinforced their neovascularization by binding to the receptor of EP4 in an AMPK-dependent manner. PGE2 may have clinical value in ischemic heart disease.

Yes but where is the science or study implying anything to do with hair regeneration? You are pointing to stem cell remodeling in wounds, this is the normal process of wound healing. There is also a risk in the remodeling process, and that is scarring alepecia, when that happens, you are essentially sealing the fate of the hair follicle for good with a hair-growth-no-more-no-jutsu (for all the naruto fans :)... The only way thing you can do to the area once occupied by the hair is neo-genesis through fgf9 based wound healing. Don't get me wrong, dermarolling is amazing, I love it, used with minoxidil it seems to work better than anything people are using today, but I haven't seen the science that it regenerate hair yet. We initially thought in this thread that we could generate significant pge2 post wound using minoxidil, but it just won't be close to what is needed for that purpose (induce fgf9 through pge2). Also I want to know if it is possible through dermarolling to reverse the fate of follicular streamers (the point of no return), as even slick looking norwood 7 have these, they are just not visible.

I see Androgen Alopecia as an endothelial dysfunction caused by local chronic inflammation. A damaged endothelium cannot function properly( imbalance between vasodilation and vasoconstriction) and EPCS cannot migrate and do their normal job to the follicles. This is probably why Minoxidil works in the first place mimicking Nitric Oxide (NO is produced by endothelial cells that acts to maintain vascular tone and increasing vasodilation). By adding the derma roller to the mix, we shifting our body focus from the chronic inflammation of the house to a rebuilding process , the endothelium (the inner lining of blood vessels) included. male pattern baldness is an impaired vascular local condition.

Not my only theory!, but maybe part of the puzzle. There is nothing wrong with your follicles, the problem is the EPC cannot migrate anymore to accomplish their functions.

Abstract

In androgenetic alopecia, overactivation of the androgen hormone cascade in genetically predisposed persons leads to miniaturization of the dermal papilla of the hair follicle and to reduction in the number of papilla cells in the scalp, but the mechanisms explaining this miniaturization have remained unclear. According to our hypothesis, the increase of dihydrotestosterone (DHT) production in the overactive androgen state inhibits cell mitosis in the dermal papilla and contributes to the induction of programmed cell death (apoptosis). Normally, DNA molecules have a negative charge, which doubles in every cell mitosis. In the catagen and telogen phases, the sulphur-rich hair moves upwards, dehydrates and develops an increasing positive charge. In a normal hair-growth cycle, the epithelial column shortens and the secondary germ is formed and it invaginates the dermal papilla by electromagnetic attraction. In the mitotic inhibition state induced by DHT, the negative charge decreases, leading to a weakening of the electromagnetic adhesion forces and weaker electrical attraction between the undifferentiated germ cells and the dermal papilla. Insulin resistance has an additional pathogenic role in the excessive miniaturization of the hair follicle. The vasoactive substances associated with endothelial dysfunction in insulin resistance induce microcirculatory disturbance, perifollicular vasoconstriction and stimulation of smooth muscle cell proliferation in the vascular wall. This leads to microvascular insufficiency and local tissue hypoxia and progressive miniaturization of hair follicles.

Abstract

The hair follicle dermal papilla which controls hair growth, is characterized in the anagen phase by a highly developed vascular network. We have demonstrated in a previous study that the expression of an angiogenic growth factor called vascular endothelial growth factor (VEGF) mRNA varied during the hair cycle. VEGF mRNA is strongly expressed in dermal papilla cells (DPC) in the anagen phase, but during the catagen and telogen phases. VEGF mRNA is less strongly expressed. This involvement of VEGF during the hair cycle allowed us to determine whether VEGF mRNA expression by DPC was regulated by minoxidil. In addition, the effect of minoxidil on VEGF protein synthesis in both cell extracts and DPC-conditioned medium, was investigated immunoenzymatically. Both VEGF mRNA and protein were significantly elevated in treated DPC compared with controls. DPC incubated with increasing minoxidil concentrations (0.2, 2, 6, 12 and 24 mumol/L) induced a dose-dependent expression of VEGF mRNA. Quantification of transcripts showed that DPC stimulated with 24 mumol/L minoxidil express six times more VEGF mRNA than controls. Similarly, VEGF protein production increases in cell extracts and conditioned media following minoxidil stimulation. These studies strongly support the likely involvement of minoxidil in the development of dermal papilla vascularization via a stimulation of VEGF expression, and support the hypothesis that minoxidil has a physiological role in maintaining a good vascularization of hair follicles in androgenetic alopecia.

What have we seen so far? From the Original study and from posters with results in this thread and other posts elsewhere like you say we know rolling is doing something positive.

But what about some of the result pics that appear to show new growth growing directly from the needle puncture sites ?

State of the situation: Day 6 after dermarolling session 4th (1.0mm needles).

Blue line highlight juvenile hairline, Pink line highlight receding hairline. Notice like some hairs are sprouting randomly in areas where I didn't have visible hair. Also, maybe it's difficult to see in this picture (phone = bad quality), notice several black dots along my receding hairline, hopefully it's new hair coming out. The white crap in my hair is dead skin, I started to peel today :)

I will use this picture for future reference.

Armando! What was your theory again?:) I am buying chemical peels online as I type this. I really think that Fibrosis is the real b**ch in Androgen Alopecia after looking at my own results with the derma roller. Hair are coming out of the punctured area only. People having success with TCA peels on the immortal forum. We almost there!
